Antigen-specific T cell immunity induced by pTRG protected the mice from the second challenge with CT-26 cells. Splenocytes were harvested from mice on the 10th day after the second CT-26 challenge. Splenocytes (1 × 106) were stimulated with 10 μg/mL of CT-26 cell lysate for 24 h. A Intracellular production of IFN-γ by CD4 and CD8 T cells. B Mean IFN-γ-producing cells among CD4 and CD8 T cells (n = 6, two-way ANOVA, mean ± SEM, **p < 0.01). C Concentrations of indicated cytokines and cytotoxic mediators in culture media were measured via enzyme-linked immunosorbent assays (ELISAs) (n = 6, two-way ANOVA, mean ± SEM, **p < 0.01). D In vivo CT-26 antigen-specific lysis was measured by CT-26 antigen-loaded splenocyte transfer. (n = 6, two-way ANOVA, mean ± SEM, **p < 0.01). E The mice were injected with anti-CD4 and CD8 Abs every 2 days from the 28th day after the first tumor challenge. The survival rate of the mice is shown (n = 5 per group)
