Intracellular cytokine production. At 10 days post-3rd immunization, the lung samples were subjected to flow cytometry analysis to evaluate the production of intracellular IFN-gamma (A), IL-4 (B), and IL-17 (C) in CD4 T cells, as well as IL-2 (D), IFN-gamma (E) and TNF-alpha (F) in CD8 T cells. The spleens were also collected, and the generation of IL-2 (G), IFN-gamma (H), TNF-alpha (I) and IL-2+ IFN-gamma+TNF-alpha+ (J) in CD8 T cells was measured. In addition, the intracellular production of IFN-gamma in the spleen was also measured by ELISpot assay (K, L)
